Linking Retroelements to Autoimmunity
In this issue, Stetson et al. (2008) report a mechanism by which host cells avert an autoimmune response to self-nucleic acids. They show that the nuclease Trex1 prevents the accumulation of DNA derived from endogenous retroelements that, if left unchecked, trigger elevated production of type I interferons leading to autoimmunity.
متن کاملBasophils and IgE: Linking the Allergic Environment to Autoimmunity
As outlined in some of the accompanying articles in this issue, the role of the basophil as an effector cell in allergy and in host defense (particularly to parasites) has long been recognized. However, recent advances advocate for the basophil as an immunomodulatory cell that can promote naïve CD4 + T cell commitment to Th2 cell differentiation. While this is in keeping with the concept that t...
متن کاملLinking autoimmunity to the origin of the adaptive immune system
In jawed vertebrates, the adaptive immune system (AIS) cooperates with the innate immune system (IIS) to protect hosts from infections. Although targeting non-self-components, the AIS also generates self-reactive antibodies which, when inadequately counter-selected, can give rise to autoimmune diseases (ADs). ADs are on the rise in western countries. Why haven't ADs been eliminated during the e...

متن کاملE2F4/5 and p107 as Smad Cofactors Linking the TGFβ Receptor to c-myc Repression
Smad3 is a direct mediator of transcriptional activation by the TGFbeta receptor. Its target genes in epithelial cells include cyclin-dependent kinase inhibitors that generate a cytostatic reponse. We defined how, in the same context, Smad3 can also mediate transcriptional repression of the growth-promoting gene c-myc. A complex containing Smad3, the transcription factors E2F4/5 and DP1, and th...
